چکیده

21 In the industrial yeast biomass production process, cells undergo an oxidative and other 22 stresses which worsens the quality of the produced biomass. The overexpression of the 23 thioredoxin codifying gene TRX2 in a wine Saccharomyces cerevisiae strain increases 24 resistance to oxidative stress and industrial biomass production yield. We observed that 25 variations in the TRX2 gene dose in wine yeast strains are relevant to determine the 26 fermentative capacity throughout the industrial biomass production process. So we 27 studied the molecular changes using a transcriptomic approach under these conditions. 28 The results provide an overview of the different metabolic pathways affected during 29 industrial biomass production by TRX2 gene manipulation. The oxidative stress-related 30 genes, like those related with the glutathione metabolism, presented outstanding 31 variations. The data also allowed us to propose new thioredoxin targets in S. cerevisiae, 32 such as Hexokinase 2, with relevance for industrial fermentation performance. 33

Metabolic engineering of malolactic wine yeast.

Malolactic fermentation is essential for the deacidification of high acid grape must. We have constructed a genetically stable industrial strain of Saccharomyces cerevisiae by integrating a linear cassette containing the Schizosaccharomyces pombe malate permease gene (mae1) and the Oenococcus oeni malolactic gene (mleA) under control of the S. cerevisiae PGK1 promoter and terminator sequences i...

Comparative transcriptomic and proteomic profiling of industrial wine yeast strains.

The geno- and phenotypic diversity of commercial Saccharomyces cerevisiae wine yeast strains provides an opportunity to apply the system-wide approaches that are reasonably well established for laboratory strains to generate insight into the functioning of complex cellular networks in industrial environments. We have previously analyzed the transcriptomes of five industrial wine yeast strains a...

Isolation, Subtype Determination, Cloning and Expression of HBsAg Gene from an Iranian Carrier in Saccharomyces cerevisiae

The Hepatitis B Surface antigen ( HBsAg) gene was isolated from an Iranian HBeAg positive carrier by PCR. The gene was cloned in pUC19 for sequencing and pYES2 for expression in Saccharomyces cerevisiae, which pNF1 and pDF3 constructs were made respectively. The sequencing data showed that the isolated HBsAg gene shared more than 90% homology with the ayw subtype. The pDF3 was transferred into ...

CLONING AND EXPRESSION OF HUMAN IFNα2B GENE IN SACCHAROMYCES CEREVISIAE

Interferon is a protein secreted by eucaryotic cells following stimulation by viruses, bacteria, and many other immunogenes. Recent medical studies indicate that interferons have effective role in the treatment of virus infections, immunodeficiency and certain types of cancer such as hairy cell leukaemia (HCL). The aim of the present study is to apply yeast strain for secreting human IFNα2b fol...

Reduction of oxidative cellular damage by overexpression of the thioredoxin TRX2 gene improves yield and quality of wine yeast dry active biomass

BACKGROUND Wine Saccharomyces cerevisiae strains, adapted to anaerobic must fermentations, suffer oxidative stress when they are grown under aerobic conditions for biomass propagation in the industrial process of active dry yeast production. Oxidative metabolism of sugars favors high biomass yields but also causes increased oxidation damage of cell components. The overexpression of the TRX2 gen...
